Town Doctor Jumbo is based on a manga of the same name. It centers around a nurse named Baba Asuka who recently started her career at the office where her father worked as a doctor. When he suddenly dies, it is up to Asuka to find a new doctor for her town and she soon becomes frantic in her search.

It is then that a man named Tsuruta Masayoshi, also known as Jumbo, shows up and buys the clinic declaring it his. He disappeared 12 years earlier and nobody knows how or why he is suddenly back or what his real motive for having bought the clinic is, but he soon proves himself as a good doctor for the town.

Kurahashi's condition is continuously getting worse and by now, only a liver transplant can save him. However, there's no time to wait for a donor liver, and the only way would be to do a living donor liver transplant. The only ones who qualify as donors are blood relatives, but Tenryuu refuses flat out to become the donor.

The hearing ends in chaos after Kurahashi plays back a voice recording of Tenryuu threatening the representative of the pharmaceutical company and revealing that Ishikura wasn't actually hurt by Jumbo. In response, Tenryuu denounces his brother and explains that Kurahashi faked the evidence out of personal spite against him.

Asuka walks in on Tenryuu beating his brother Kurahashi. Tenryuu then reveals to her that Kurahashi had just been toying with her to infiltrate the Baba Clinic. Kurahashi confirms this as a fact and crushes any hope Asuka might have had.

Based on Tenryuu's suggestion, Kurahashi and Asuka start their new life together away from the Baba Clinic. However, Kurahashi's old teacher tries to call on him and suddenly collapses with his left side paralyzed. With all hospitals without capacity on this holiday, Kurahashi has no choice but to take him to the Baba Clinic.

Asuka finds her musician friend Momota Tetsuo drunk by the sea. They take him to Baba Clinic to check him out, but then things are more serious than initially thought. He's diagnosed with Brinton's disease, a type of stomach cancer.

A patient with lung cancer in its terminal stage escapes from Shirane General Hospital. While the staff is scrambling to find him, Jumbo runs into him as he's peeking into the Baba Clinic, where Asuka is throwing a party for her soon to be married friend Haruna.

Jumbo and Asuka rush to the harbor in response to an emergency call. But when they get there, nothing's really wrong. Just when they are about the leave, however, a real emergency unfolds as a boy tries to commit suicide.

Asuka, acting on her father's words, tries to help two local children who stopped by the clinic. The children's parents don't appreciate this and concoct a story of Asuka abusing them, in order to extort money from the clinic. All the while the little girl's medical condition goes undiagnosed and begins to worsen.
